Use | 1400W is a highly selective inducible nitric oxide synthase inhibitor and is a potential disease modifier in the rat kainate model of temporal lobe epilepsy. 1400W reduces ischemia reperfusion injury in an ex-vivo porcine model of the donation after circulatory death kidney donor. 1400W ameliorates acute hypobaric hypoxia/reoxygenation-induced cognitive deficits by suppressing the induction of inducible nitric oxide synthase in rat cerebral cortex microglia. 1400W blocks death pathway of LPS-induced activated-microglia to preOLs. |
In vitro study | 1400W is an irreversible inhibitor or an inhibitor with positive and slow reversibility, inhibiting iNOS. 1400 inhibition of human iNOS was time-dependent. 1400W and L-arginine belong to the competition relationship, not the substrate of iNOS. |
In vivo study | 1400W selectively inhibited microvasculature injury in rats. In a rat model of endotoxin-induced vascular injury, 1400W was more than 50 times more potent at inhibiting iNOS than eNOS. In addition, 1400W also concentration-dependently reduced LPS-induced vascular leakage associated with iNOS induction in the colon, liver, kidney, and heart. For the removal of other organs of the kidney, its protective effect is almost 100%, while in the kidney 54%. 1400W in the rat model of renal ischemia reperfusion, the oxidation and nitrification pressure of the kidney were improved. In mice, treatment with 1400W can reduce the growth rate of solid tumors. |
